La forme d'un aptam\u00e8re est tr\u00e8s importante pour son affinit\u00e9 de liaison \u00e0 la cible. Cela est d\u00fb au fait que la forme lui permet de \"s'adapter\" \u00e0 une cible, mais oriente \u00e9galement les nucl\u00e9otides impliqu\u00e9s dans la liaison de mani\u00e8re \u00e0 optimiser les interactions de liaison. Pour que les aptam\u00e8res soient d\u00e9velopp\u00e9s pour des produits commerciaux, ils doivent constamment \u00eatre dans la forme id\u00e9ale pour une liaison efficace. Cela signifie qu'une optimisation est n\u00e9cessaire.<\/span><\/p>\n

L'optimisation est n\u00e9cessaire car, m\u00eame si les aptam\u00e8res sont toujours repr\u00e9sent\u00e9s dans une confirmation de forme unique, leur forme fluctue constamment \u00e0 temp\u00e9rature ambiante. Cela peut cr\u00e9er un probl\u00e8me parce que si l'aptam\u00e8re est dans une forme de liaison d\u00e9favorable lorsqu'il entre en contact avec une cible, il ne se liera pas. Pour am\u00e9liorer la liaison, l'aptam\u00e8re doit \u00eatre le plus souvent possible dans la forme qui permet la liaison. Nous utilisons la mod\u00e9lisation informatique de structures secondaires pr\u00e9dites en flux comme base pour la truncation et l'optimisation des aptam\u00e8res.\u00a0<\/span><\/p>\n
