Typiquement, cela est accompli en immobilisant la cible de sorte que les aptam\u00e8res qui se lient restent attach\u00e9s, tandis que ceux qui ne se lient pas sont \u00e9limin\u00e9s par lavage. Le grand inconv\u00e9nient de l'immobilisation des cibles est que partie de la cible est retir\u00e9e de la s\u00e9lection du fait qu'elle est attach\u00e9e au support solide. Cela pose probl\u00e8me avec chaque cible, des petites mol\u00e9cules (supprimant enti\u00e8rement les \u00e9pitopes pour la liaison) aux grandes prot\u00e9ines (pouvant affecter le pliage). Nous avons pu r\u00e9soudre ce probl\u00e8me en d\u00e9veloppant la plateforme FRELEX, qui permet de s\u00e9parer les aptam\u00e8res li\u00e9s \u00e0 la cible de ceux qui ne le sont pas d'une mani\u00e8re o\u00f9 ni la cible ni l'aptam\u00e8re ne sont immobilis\u00e9s sur une surface.<\/p>\n<p>&nbsp;<\/p>\n<h3>Qu'est-ce que FRELEX ?<\/h3>\n<p>La plateforme FRELEX utilise une biblioth\u00e8que d'oligonucl\u00e9otides courts al\u00e9atoires sur une puce d'or. Ces oligonucl\u00e9otides courts agissent comme une \u00ab pelouse \u00bb sur laquelle les aptam\u00e8res peuvent \u00ab s'asseoir \u00bb par hybridation via l'appariement Watson-Crick. Nous s\u00e9lectionnons d'abord les s\u00e9quences d'aptam\u00e8res qui s'assoient sur cette pelouse sans que la cible soit pr\u00e9sente. Les s\u00e9quences qui se sont li\u00e9es \u00e0 la surface sont retir\u00e9es, puis incub\u00e9es avec la cible et r\u00e9introduites sur la pelouse de la puce d'or.
