Les aptam\u00e8res sont des oligonucl\u00e9otides \u00e0 simple brin (ARN ou ADN). Ils sont compos\u00e9s de nucl\u00e9otides, pas d'acides amin\u00e9s. Les oligonucl\u00e9otides \u00e0 simple brin cherchent \u00e0 \u00eatre le plus double brin possible, donc les r\u00e9gions d'homologie interne se replient sur elles-m\u00eames formant une forme 3D. C'est en gros la m\u00eame chose que pour les anticorps, les aptam\u00e8res se lient \u00e0 d'autres mol\u00e9cules en fonction de cette forme, et de l'arrangement 3D des charges pr\u00e9sent\u00e9 par la forme.\u00a0<\/span><\/p>\n<p><span style=\"font-weight: 400;\">L'identification des aptam\u00e8res est diff\u00e9rente de celle des anticorps. Au lieu de commencer avec un r\u00e9pertoire na\u00eff relativement petit, nous synth\u00e9tisons un grand ensemble (un quadrillion de s\u00e9quences diff\u00e9rentes) et exposons toutes ces s\u00e9quences \u00e0 la cible. Les aptam\u00e8res sont synth\u00e9tis\u00e9s sur des instruments, ils ne sont pas d\u00e9riv\u00e9s de cellules vivantes. Ces ensembles d'aptam\u00e8res (biblioth\u00e8ques) sont expos\u00e9s \u00e0 une cible d\u00e9finie en laboratoire et les s\u00e9quences qui se lient \u00e0 la cible sont conserv\u00e9es, tandis que celles qui ne se lient pas sont \u00e9limin\u00e9es. Les s\u00e9quences conserv\u00e9es sont amplifi\u00e9es par PCR et le processus est r\u00e9p\u00e9t\u00e9 plusieurs fois. Les biblioth\u00e8ques s\u00e9lectionn\u00e9es sont ensuite caract\u00e9ris\u00e9es par s\u00e9quen\u00e7age de nouvelle g\u00e9n\u00e9ration (NGS) et les s\u00e9quences ayant le nombre de copies le plus \u00e9lev\u00e9 sont identifi\u00e9es. Celles-ci sont synth\u00e9tis\u00e9es puis test\u00e9es, \u00e0 nouveau in vitro, pour leur capacit\u00e9 \u00e0 se lier \u00e0 la mol\u00e9cule cible.\u00a0<\/span><\/p>\n<p>&nbsp;<\/p>\n<p><span style=\"font-weight: 400;\">Ainsi, le principe de fonctionnement des aptam\u00e8res est le m\u00eame que celui des anticorps : ils forment des structures qui se lient \u00e0 n'importe quelle mol\u00e9cule cible. Le m\u00e9canisme de liaison est le m\u00eame, \u00e0 savoir les forces de Van der Waals et les liaisons hydrog\u00e8ne, celles-ci \u00e9tant simplement form\u00e9es entre les nucl\u00e9otides et la cible dans le cas des aptam\u00e8res, contre les acides amin\u00e9s dans le cas des anticorps.<\/span><\/p>\n<p><span style=\"font-weight: 400;\">Ainsi, tout comme les cow-boys et les extraterrestres sont tous deux des formes qui peuvent faire des choses similaires mais sont constitu\u00e9s de composants diff\u00e9rents, les aptam\u00e8res et les anticorps sont des formes qui font des choses similaires.
